Human experience has demonstrated redness, cracking and blistering of skin through repeated and prolonged contact with N-Methyl pyrrolidone. NMP is reported to be fetotoxic and teratogenic by dermal exposure. Rats exposed to an aerosol-vapor mixture of NMP at 1.0 mg/l for 6 hr/day, 5 days/week for 4 weeks developed excessive mortality, focal pneumonia, bone marrow hypoplasia and atrophy of lymphoid tissue. Teratogenic effects: Male and female rats exposed orally to 500 mg/kg/day in feed for 10 weeks before mating and throughout pregnancy: fetolethality. In earlier study, 300 mg/kg/day was a clear no effect level.
